Job Summary

Post-doctoral fellowship to characterize the functional impact of TWIST1 dimerization motifs on malignant phenotypes of glioma stem cells

A post-doctoral fellow is sought to join the Rostomily lab at the Houston Methodist Research Institute.  The fellow will participate in the study of TWIST1 dimerization motifs and the molecular mechanisms by which they influence glioma stem cell (GSC) phenotypes.  TWIST1, a bHLH transcription factor (TF), is a central regulator of epithelial mesenchymal transition (EMT) in a wide variety of cancers.  In glioma we have shown that TWIST1 promotes invasion, tumorigenicity and stem cell properties.  TWIST1 transcriptional activity requires the formation of homo- or heterodimers.  The current project seeks to determine how specific TWIST1 dimer motifs influence malignant properties of GSCs, identify mechanisms which govern the formation of specific dimer motifs and elucidate dimer motif-specific downstream molecular mechanisms which regulate GSC malignant properties.  Given the difficulties in targeting TFs directly, these studies are expected to provide new insights into alternative therapeutic strategies to indirectly target TWIST1 function through TWIST1 regulated proteins.  The candidate must have a strong background in molecular and cellular cancer biology and expertise in a variety of the approaches utilized for this project including cloning and characterization of tethered protein-protein expression constructs, RNAseq, CHIPseq, promoter reporter assays, proteomics, CRIPSR or shRNA mediated loss of function, GSC culture and generation and immuno-histologic characterization of xenograft tumors. Experience with bioinformatics tools for data analysis is a plus.
